The Null Allele Polymorphism of 17 Y-STR loci in Thai Population

Abstract

In routine paternal testing using Y-chromosomal short tandem repeat (Y-STR) analysis, when allele dropout or null alleles are observed at certain loci and it is not possible to determine conclusively whether this phenomenon arises from laboratory techniques or the sample itself, time consuming and additional expense are required to obtain accurate results. In this current report, the data from routine casework were analyzed. A study of 400 male individuals in Thai population using Y-STR in routine service of Human Genetic Laboratory during 2017-2020 revealed distinct Y-STR haplotypes associated with null alleles. Genotyping of these samples was performed at 17 Y-STR loci utilizing AmpFlSTR Yfiler kit. We observed allele dropout in six cases, two from the DYS456 locus and four from the DYS448 locus. Our results showed that DYS456 and DYS448 loci exhibited such mutations at frequencies of 0.5% and 1%, respectively. The current data provided useful information on null alleles for forensic DNA laboratories in Thailand involving Y-STR analysis in paternal testing, reducing discrepancy error and forensic casework investigation.
