Complement C3 level and expression of CD55 and CD59 on red blood cells of variable clinical severity Hb H disease

Abstract
This is the first study which aimed to determine the complement C3 level and the expression of CD55 and CD59 on red blood cells of variable clinical severity Hb H patients. This study conducted on 44 patients from Sunpasitthiprasong Hospital, Ubon Ratchathani, Thailand. Blood sample were collected 30 days after being transfused or just before receiving a transfusion. Measurement of complement C3 level was done using turbidimetric test. Investigation of CD55 and CD59 expression on red blood cell was performed using flow cytometry method. Among these patients, 14 patients were categorized as mild phenotype, 6 patients were moderate and 24 patients were severe phenotype. The results showed that complement C3 level in mild, moderate and severe Hb H disease were 1.08 ± 0.16, 1.15 ± 0.29 and 1.05 ± 0.17 g/L, respectively. The expression of CD55 in mild, moderate and severe type were 87.4 ± 12.22, 77.8 ± 14.94 and 95.8 ± 3.42 %, respectively. The expression of CD59 in those groups were 98.3 ± 1.89, 97.6 ± 2.23 and 99.0 ± 1.48 %, respectively. Statistical analysis showed no significant difference in the level of CD55 and CD59 expression in patients with variable severity and control group. However, this study revealed the trend of low level of CD55 in more severe phenotype of Hb H disease. The further study of these molecules might give the insight understanding about CD55 and CD59 expression on variable clinical severity Hb H disease.
