Abstract

Environmental surveillance for SARS-CoV-2 contamination is a crucial strategy for preventing and controlling nosocomial infections. This study aimed to evaluate the effectiveness of two sampling techniques for detecting SARS-CoV-2 contamination: The Dual-Swab Pooled Sampling technique and the Individual Swab Sampling technique. Samples were collected frequently on high-touch surfaces such as door handles, handrails, faucets, and medical equipment within urban healthcare units. Samples were collected between August and October 2022. The samples were then examined in the laboratory for the Orf1ab, N, and E genes, detected by using Real-time RT-PCR combined with the Cycle Threshold (Ct) value.

The results found a total of 79 samples; the genetic sequence of the SARS-CoV-2 virus was detected in 4 samples (5.06%). The highest levels of viral contamination were found in the male’s restroom on the 1st floor, specifically on the door lock (Ct 32.82–34.50) and the spray handle (Ct 30.51–31.17). Additionally, genetic material of the SARS-CoV-2 virus was found on the female’s restroom sink faucet on floor 4, and the entrance door handle on floor 5. The efficiency of the Dual-Swab Pooled Sampling technique for high-touch surfaces achieved a sensitivity of 100% compared to the Individual Swab Sampling technique. The average difference in Ct values ​​(ΔCt) between the two methods was 0.8 ± 0.3, with a p-value > 0.05, indicating no statistically significant difference. The study results indicate that the Dual-Swab Pooled Sampling technique is effective, preventing sample dilution even at low viral loads. Furthermore, it reduces laboratory workload by approximately 50% and can be applied to implement measures to prevent the spread of SARS-CoV-2 in the environment of healthcare units.
