Factor VIII: C and von Willebrand Factor Level in Heat Treated Freeze Dried Cryoprecipitate

Abstract: The National Red Cross Blood Center in Thailand has produced Heat Treated Freeze Dried Cryoprecipitate (HTFDC) to replace fresh frozen plasma and frozen cryoprecipitate for the treatment of Hemophilia A and von Willebrand disease. HTFDC is prepared from 3 unit pooled frozen cryoprecipitate in glycine buffer, then lyophilized and heat treated at 60 C for 72 hours. It is stable at 4 C, which is convenient for transportation, storage and infusion. One bottle of HTFDC is dissolved in 50 ml. sterile water, with protein cont of 25 g/bottle. The levels of factor VIII clotting activity (FVII:C), von Willebrand for antigen (vWF:Ag) and ristocetin cofactor activity RiCoF) of HTFDC in 69 bottles were studied by one stage clotting assay, ELISA and agglutination respectively. The mean levels of FVIII C, vWF: Ag and RiCoF in one bottle of HTFDC were 336 41 108.9,655:5+163 2 and 6544+312.9 units respectively. The ressults showed that the locally prepared HTFDC is suitable for the treatment in patient with Hemophilia A and vWD. It has been effectively used in controlling bleeding episodes in the hospital as well as home care therapy.
