DNA Extraction from Buffy Coat Sent by Mail Without Ice

Abstract: DMA was extracted from 22 samples of butfy coat sent by mail without ice from eight different provincial and university hospitals in Thailand. The transportation took one to five days. The results revealed that 16 samples (73%) of buffy coat could be used for DNA extraction and six samples (27%) were ungualified due to clotting (n = 5) and only plasma was obtained (n = 1). By extracting DMA from 500 Ul. of buffy coat, it yielded the mean ratio of OD 260/OD 280 of 1.56 and the mean amount of DNA of 68 plg. Therefore, we conclude that the buffy coat sent by mail without ice is suitable for extracting DNA The vieided DMA can be used for further studies. It is simple, practical, efficient and of low cost.
