Introduction: The mixture of Phet Sang-Khaat capsule is recommended to treat hemorrhoid in Thai National List of Essential Herbal Medicines (2011). This formula consists of 3 medicinal plants including Cissus quadrangularis (Linn.) (Vitaceae), Clerodendrum serratum (L.) Moon. var. wallichii C.B.Clarke (Lamiaceae) and Rheum officinale Baillon. (Polygonaceae). The previous review literatures on the mixture of Phet Sang-Khaat capsule did not contain information about quality control or formulation. However, we found several phytochemical and pharmacological studies on single ingredients of this formula. The aim of the study to determine the contents of some active constituent in each ingredient of formulation such as quercetin in C. quadrangularis and rhein in R. officinale and the mixture of Phet Sang-Khaat capsule.  Methods: The dried powder of the samples were extracted with 50% ethanol. HPLC was used to determine each of medicinal plants preparation content of major compounds from these 3 medicinal plants from 5 different regions. Results: The results showed that the amount of quercetin in C. quadrangularis from Khon Kaen, Nakornratchasima, Maha Sarakham, Phetchabun and Roied provinces were 1.8, 1,69, 3.51, 3.15 and 3.30 (%w/w, dried powder), respectively. The amount of rhein in rhubarb from Bangkok, Khon Kaen, Maha Sarakham, Nakornratchasima, and Roied provinces were 1.02, 1.09, 1.01, 0.54 and 1.56 (%w/w, dried powder), respectively. Thus, we were used the aerial part of C. quadrangularis from Maha Sarakham and the rhizomes of rhubarb from Roied province as the ingredients to formulate the mixture of Phet Sang-Khaat capsule in this study. The amount of querceitn and rhein in the mixture of Phet Sang-Khaat capsule were obtained 10.96 and 0.70 mg/capsule, respectively. Conclusion: The results from this study could be used as a guideline for the quality of some Thai herbal formulation.
